Gene therapy is a promising strategy for many neurodegenerative disorders such as Parkinson's disease, Alzheimers disease or Huntington's disease. The general idea behind gene therapy is to replace or silence faulty genes in order to correct the disease phenotype. Gene therapy is based on two main technologies, the first being the gene-editing mechanism itself whereas the second is based on the vehicle to deliver the gene-editing mechanism to the cell. The current project will work on the latter. Traditionally, viral vectors have been/are used to deliver the cargo to the target cell. These vectors are taken from nature and modified to be replication deficient. The naturally evolved vector variants do not have a high specificity (tropism) for every cell type. We have recently generated a large libary of AAV vector capsids (the virus shell) that we have injected into the brain of rodents. In the proposed project the student will analyse which different vectors target certain cell populations efficiently (ie. astrocytes, neurons, microglia, etc.).

The project will involve immunohistochemistry, microscopy, and cell quantification methods.
