Description: 

To investigate the influence of mitochondrial DNA (mtDNA) replication on neuronal differentiation of induced Pluripotent Stem Cells expressing genes that cause familial Alzheimer's disease. Our laboratory has recently developed a digital PCR method called Selfie-Digital-PCR that allows obtaining absolute measurements of mtDNA copies and mitochondrial RNA transcripts. With Selfie-dPCR we investigate the replication / transcription of mitochondrial DNA in stem cells and neural precursors during their differentiation process. By participating in this project, the trainee will gain experience in a wide range of techniques that include recombinant DNA techniques for the design and packaging of viral vectors, cell culture, digital PCR, confocal spinning disk microscopy in live cells, as wells as analysis and discussion of results.
